2371-ben egy csillagközi anomáliának, a Vadvidéknek köszönhetően egy maquis hajó és az azt üldöző Voyager átkerül a Galaxis túlsó végébe, a Delta kvadránsba, ahol egy Gondviselő nevű magasabbrendű lénynek szüksége van a segítségükre. A súlyos károsodást szenvedett csillaghajón az első tiszt, a kormányos, a főgépész és a teljes orvosi személyzet is életét veszti. A Gondviselő haldoklik, viszont nem akarja magára hagyni a rábízott bolygó népét, az ocampákat, akik még nem elég felkészültek a planéta elhagyására vagy az agresszív kazonok elleni védekezésre. A Voyager feladata, hogy átsegítse az ocampákat a Gondviselő elvesztésén, illetve hogy elpusztítsa a Gondviselő otthonaként szolgáló űrállomást, amit az ocampák ellen lehetne fordítani. Ezzel viszont elvágják az azonnali hazafelé vezető utat is, de Janeway kapitány fontosabbnak ítéli egy nép létbiztonságát a saját érdeküknél. Még itt a hajóra kerül a talaxiai Neelix és barátnője, az ocampa Kes: előbbi a Voyager szakácsa, utóbbi pedig a Doktor asszisztense lesz. Az üldözött maquis hajó legénysége a Voyager fedélzetére kerül; Chakotay, a maquis hajó vezetője a Voyager első tisztje (parancsnoka) és B’Elanna Torres (egy félig ember, félig klingon) gépész pedig a Voyager főgépésze lesz. Tuvok (vulkáni), aki a Föderáció kémje volt a maquis soraiban, a Voyager biztonsági főnöki posztját tölti be. Tom Paris, aki eddig csak megfigyelő volt a hajón, ideiglenesen hadnagyi rangot kap, és a főkormányosi széket foglalja el, az orvosi főtiszt helyébe pedig a Orvosi Segédhologram (OSH) lép. Később Kes távozik a hajóról, míg egy borg dolgozó, Hét Kilenced, Hetes érkezik a hajóra. A kapitány mindenben segíti Hetest az emberré válás folyamatában. Így kezdődik a Voyager hosszú, kalandokkal szegélyezett útja az Alfa Kvadráns irányába, amit elvileg 75 év alatt érhetnek el. Janeway abban reménykedik, hogy találnak egy a Gondviselőhöz hasonló lényt, aki segíthet őket hazajuttatni.Legelőszőr 2374-ben kapnak leveleket sz Alfa Kvadránsban maradt hozzátartozóiktól , majd 2376-ban sikerül elsőnek hang kapcsolatot létrehozniuk a Csillagflotta Parancsnoksággal , de aztán videó kapcsolatot is rendszeresen létre tudnak hozni . A Voyager kalandjai szintén nem kifejezetten különálló részekben élvezhetőek, érdemes végigkövetni a szereplők (külső-)belső alakulását a közösen átélt viszontagságok során. A sorozatnak némi feminin jelleget ad, hogy a Star Trek produkciók közül itt legtöbb aktív női szereplő: a kapitányon, Janeway-en kívül több fontos női karakter, Torres, Kes, Hét Kilenced és később még a Borg-királynő is jelentős alakítói a cselekménynek.
